Rooting and acclimatization of micropropagatedHypericum perforatumL. native to Sicily

2017

Hypericum perforatum L. is traditionally used as a medicinal plant because of its different bioactive compounds with documented antidepressant and antiinflammatory activities. Plantlets’ mass production with high content of these secondary metabolites has been enhanced through in vitro culture but the process has often been stopped at the multiplication phase. A study was conducted in order to set up an efficient in vitro rooting and acclimatization protocol of a H. perforatum Sicilian genotype well-adapted to south Mediterranean conditions. Aseptic nodal segments were cultured onto a Murashige and Skoog (MS) basal medium supplemented with 4.44 µM 6-benzyladenine for multiplication. Microsh…

In vitro response of two Sicilian genotypes of Morus (L.) through axillary bud culture.

2007

A protocol for the in vitro establishment of two Sicilian genotypes of mulberry has been set up. Multiple shoots were initiated and plantlets were produced from buds collected from mature trees of Morus alba L. and Morus nigra L., grown in the field. The influence of two explanting times (September and December) of buds on the in vitro response has been observed. The percentage of sprouting after four weeks of culture was greatly higher with material explanted from the field in September compared with the one collected in December. Morus nigra L. produced more vigorous shoots than Morus alba L.. After the second subculture the number of shoots produced decreased, in both genotypes. No diffe…

Selection and micropropagation of valuable caper genotypes

2020

The high quality of the various biotypes present in the natural or cultivated state is one of the main features for caper production. Up to now, however, no selection activity has been carried out in order to identify the most suitable accessions for providing better quality products. In this paper, we report the first results of a selection of caper genotypes characterised by significant qualitative traits. A micropropagation protocol was evaluated in order to improve and allow the multiplication of the most promising Capparis spinosa L. subsp. spinosa genotypes, selected in Salina (Aeolian Islands), Sicily (Italy), in collaboration with the most important local growers.

Somatic embryogenesis in Abies nebrodensis, an endangered Sicilian fir

2022

AbstractSomatic embryogenesis, as a promising biotechnological tool for many conifer trees, has never been applied for the Abies nebrodensis species. Although all the encouraging results previously obtained by the EU LIFE (European LIFE program) funded projects in over ten years, the critically endangered Sicilian fir remains alarmingly close to extinction. In this study, we reported the first protocol of somatic embryogenesis obtained from mature zygotic embryos of the Abies nebrodensis. Seeds from Abies adult trees with specific identification numbers (IN) were collected and full seeds were identified by X-ray. Different experiments were carried out for callus initiation, from both zygoti…

Primary culture of avian embryonic heart forming region cells to study the regulation of vertebrate early heart morphogenesis by vitamin A

2014

Background: Important knowledge about the role of vitamin A in vertebrate heart development has been obtained using the vitamin A-deficient avian in ovo model which enables the in vivo examination of very early stages of vertebrate heart morphogenesis. These studies have revealed the critical role of the vitamin A-active form, retinoic acid (RA) in the regulation of several developmental genes, including the important growth regulatory factor, transforming growth factor-beta2 (TGFβ2), involved in early events of heart morphogenesis. However, this in ovo model is not readily available for elucidating details of molecular mechanisms determining RA activity, thus limiting further examination o…

Analisi delle popolazioni siciliane di Helichrysum: propagazione in vitro e indagini fitochimiche

2004

The Sicilian populations of Helichisum have got a germoplasm from which it is possible to select ornamental plants to be placed in a mediterranean and/or dry climate. The in vitro trials show an attitude for microprapagation and multiplication for commerciaI productions that can be used also for the re·introduction in the natural habitat. The highest multiplication rate belongs to three genotypes: H. hyblaeum (clone S6), H. nebrodense (Clone Cli) and H. rupestre var. rupestre (clone S2, S11). The height and the feature of the vitro plants are pecuIiar for each different clone. The clone S11 is suitable for the production of pot plants or bushes, the clone S2 and S6 for garden edge. The clon…
